Medical Biotechnology Laboratory (MBL)

Introduction

Established as a separate unit in 2005, the Medical Biotechnology Laboratory (MBL) is located within the Faculty of Medicine, University of Malaya. The laboratory comprises four major sections, the Proteomics facility, Genomics facility, Bioinformatics facility and a Clean Room. The laboratory, spanning over a 60,000 sq. ft area is designated as a centralised research facility housing the Faculty’s high technology and basic equipment for conducting researches.

In line with University of Malaya’s vision, MBL’s vision is to be a premier research laboratory, providing a well equipped laboratory for the advancement of medical biotechnological research at the Faculty of Medicine and in the University of Malaya.

The main mission of MBL is to integrate collaboration for research excellence and to establish core facilities to meet the needs of advanced medical biotechnological research.

The prerequisites for MBL was previously supported by the National Biotechnology Directorate, Ministry of Science, Technology and Environment (MOSTE) for the facilities’ equipment, together with the basic laboratory inventories. At present MBL is fully supported by University of Malaya’s own grant.

As a centralised facility, MBL is made available for the researchers in University of Malaya and Malaysian scientists in the field of proteomics, genomics, bioinformatics and tissue culture. The main equipment provided are: MALDI MS, Tempo LC-MALDI, SELDI-ToF ProteinChip System, Biacore SPR, Real Time PCR, Multiplex ELISA System (BIOPLEX) and EttanDalt 2DE system. MBL also provide services for analysis of samples and serve as training centre for researchers in the respective areas.

Central Research Equipment

Charged services are offered for sample analysis and use of the following equipment:

1. MALDI TOF/TOF (ABI 4800 Plus) and TEMPO MALDI-LC

Location       :

Proteomics Facility , Medical Biotechnology Laboratory

Description :

Mass spectrometry has assumed a key role for the analysis and identification of proteins in the modern proteomics workflow. The MALDI TOF/TOF instrument is mainly used for accurate molecular mass determination of peptides and unambiguous identification of proteins.

Typical applications and uses:

  • Proteins MW measurements ranging 10 kDa and up to 300 kDa.
  • Protein identification by Peptide Mass Fingerprint (PMF) and tandem MS analysis.
  • De novo sequencing.
  • In conjunction with the adequate sample preparation pipeline: peptide and protein profiling.
  • Mass spectrometric read-out for kinetic enzymatic studies.
  • Post translational modifications (PTM) characterization.
  • Stable isotope based relative quantitation.

2. PROTEIN CHIP SELDI SYSTEM (PBS II, Biorad)

Location       :

UM Centre for Proteomics Research ( UMCPR) , Medical Biotechnology

Description : 

The system uses surface-enhanced laser desorption/ionization (SELDI) technology which combines two powerful techniques: chromatography and mass spectrometry.  The system integrates ProteinChip arrays, a SELDI reader and SELDI software to analyze samples based on measured time-of-flight (TOF).

Typical applications and uses:

  • he system will be mainly used in protein  profiling and biomarker discovery.

3. BIACORE 3000 (GE Healthcare)

Location       :

UM Centre for Proteomics Research ( UMCPR) , Medical Biotechnology

Description : 

The BIAcore 3000 system is an instrument that uses SPR (Surface Plasmon Resonance) technology for measuring the interaction of macromolecules with each other, and with small ligands.  One of the ligands is immobilized on carboxymethylated dextran over a gold surface, while the second partner (analyte) is captured as it flows over the immobilized ligand surface. Interaction is detected via SPR, in real time, at high sensitivity, without the use of radioactivity.

Typical applications and uses :

  • The BIAcore can be used for measuring binding constants of macromolecular interactions (protein-protein, nucleic acids-proteins, protein ligand ), epitope mapping, ranking antibody affinities, concentration measurements, characterization of mutant proteins and ligand fishing.

4. HIGH CONTENT SCREENING (Cellomics, Inc./VTi Array Scan)

Location       :

Centre for Cell Biology and Drug Discovery, Department of Pharmacology.

Description : 

ArrayScan VTI HCS Reader is a modular High Content Screening instrument designed for high-capacity automated fluorescence imaging and quantitative analysis of fixed and live cells. The instrument can be utilised for rapid and high content cellular screening of the bioactivity and toxicology of drugs and natural products including traditional herbal medicines, as well as analysis of chemical and biological compound.
